I have a problem with Dissolution of suspension. If I pour sample before start the paddle I observe that the release is greater than that of pouring the sample after starting the paddle. This is the observe fact.
What is the actual stratergy for Dissolution of Suspension wheather the sample should pour in the vessel before starting the paddle or after starting the paddle?

as u have mentioned that u r getting more release before putting the sample before starting the paddle,may be due to the more centrallised solubility nearby the paddle, whereas it is opposite.
Or it may be as u r putting the sample before operating paddle, there is a chace of before solubility as u wll take some time for putting sample and after that u will operate the paddle, which is not the case with putting sample with stirring.

Are we suppose to performed dissolution on suspension ? I have never seen this kind of experiment in USP or BP. Have you some reference about that ? Do someone have seen a guideline asking for that ?

I do not know if is an official requirement in the USP to perform Dissolution on suspensions. However quite a few years back it was request of the company that I was working for at the time to develop a method of the dissolution of suspension that we had on the market. At the time there were rumors that it woulfd be made compendial.
