dear friends,
i am working in a formulation research foundation.
i am working on colon Specfic drug delivery systems
how should i carry out the dissolution of my tablet formulation
i think initial 2 hrs in 0.1 N HCl
4 hs in Phosphate Buffer 7.4
remaining in Phosphate buffer 6.0
am i right?

I would agree with the plan.

There was an article on this published this spring but I can't find the reference. I think the article used less time in the HCl and a slightly lower pH for the final step.

The duration of testing in different media appears to be logical. Depending on the technology used to make colon-specific formulation (pH dependent, time dependent or enzymatic degradation by colonic flora) you may have to add enzymes to the pH 6.0 buffer. The volume of pH 6.0 buffer is also important as the amount of colonic fliud is limited.
